According to Joel Whitburn, it was issued only on 12".

As far as commercial single formats, it's my understanding that "U Can't Touch This" was only made available on 12" vinyl. By 1990, few consumers were still purchasing vinyl singles and this explains why the song's #8 peak on the Billboard Hot 100 was not truly indicative of the single's enormous popularity at the time.

Well, it could have been issued as a cassette single -- I mean, it's not like Whitburn hasn't been wrong before -- but I don't see any listing for it on GEMM as a cassette single at all. The original chart could be wrong, or it could have been scheduled for release as a cassingle and then canceled.
